Compound 1 inhibited phospholipid metabolism and Epstein-Barr virus activation caused by a potent tumor promoter. In an in vivo experiment, topical application of 1 suppressed skin-tumor-formation induced by 12-O-tetradecanoylphorbol-13-acetate (TPA) in 7,12-dimethylbenz[a]anthracene (DMBA) initiated mice. And it also suppressed ornithine decarboxylase activity stimulated by TPA on mouse skin. These results indicated that 7-isopentenyloxycoumarin is one of the effective compounds from natural resources for treating skin tumor formation.  (+info)

Allelic variation in the Depressaria pastinacella CYP6AB3 protein enhances metabolism of plant allelochemicals by altering a proximal surface residue and potential interactions with cytochrome P450 reductase. (4/8)

CYP6AB3v1, a cytochrome P450 monooxygenase in Depressaria pastinacella (parsnip webworm), is highly specialized for metabolizing imperatorin, a toxic furanocoumarin in the apiaceous host plants of this insect. Cloning and heterologous expression of CYP6AB3v2, an allelic variant identified in D. pastinacella, reveals that it metabolizes imperatorin at a rate (V(max) of 10.02 pmol/min/pmol of cytochrome P450 monooxygenase (P450)) significantly higher than CYP6AB3v1 (V(max) of 2.41 pmol/min/pmol) when supplemented with even low levels of cytochrome P450 reductase. Comparisons of the NADPH consumption rates for these variants indicate that CYP6AB3v2 utilizes this electron source at a faster rate than does CYP6AB3v1. Molecular modeling of the five amino acid differences between these variants and their potential interactions with P450 reductase suggests that replacement of Val(92) on the proximal face of CYP6AB3v1 with Ala(92) in CYP6AB3v2 affects interactions with P450 reductase so as to enhance its catalytic activity. Allelic variation at this locus potentially allows D. pastinacella to adapt to both intraspecific and interspecific variation in imperatorin concentrations in its host plants.  (+info)

Heracleum Sosnowskyi was discovered in 1772 and described as a separate species in 1944 by I. P. Mandenova. Its name is derived from the surname of a botanist studying Caucasian flora, Prof. D.I. Sosnowski. In the area of the Caucasus foothills, the plant reaches approximately 1-1,5 m in height, whereas in Poland its size is significantly larger, up to 3-3.5 m. Heracleum blooms from mid-June to the end of July. The flowers are arranged in umbels and last for 2-3 weeks. In Central Europe, the species colonizes mostly neglected green areas, ruins and riversides. Heracleum poses a serious threat to the human population due to its photoallergic properties, resulting from the presence of intensely toxic furanocoumarin in its sap. Furanocoumarins are found in small hairs that cover the leaves and stem, and are the components of the essential oil. They may penetrate the skin through the epithelial layer, posing a direct threat to human health. Contact with the plant, followed by sun exposure, may lead to the development of large blisters and symptoms of burns. Heracleum, in the event of consumption, is also harmful to farm animals, causing, among others, internal bleeding and diarrhea.
