Characterization of an acyltransferase capable of synthesizing benzylbenzoate and other volatile esters in flowers and damaged leaves of Clarkia breweri. (1/13)

A cDNA encoding a protein with 456 amino acids whose sequence shows considerable similarity to plant acyltransferases was identified among 750 Clarkia breweri flower expressed sequence tags. The cDNA was expressed in Escherichia coli, and the protein produced was shown to encode the enzyme benzoyl-coenzyme A (CoA):benzyl alcohol benzoyl transferase (BEBT). BEBT catalyzes the formation of benzylbenzoate, a minor constituent of the C. breweri floral aroma, but it also has activity with a number of other alcohols and acyl CoAs. The BEBT gene is expressed in different parts of the flowers with maximal RNA transcript levels in the stigma, and no expression was observed in the leaves under normal conditions. However, BEBT expression was induced in damaged leaves, reaching a maximum 6 h after damage occurred. We also show here that a closely related tobacco (Nicotiana tabacum) gene previously shown to be induced in leaves after being challenged by phytopathogenic bacteria also has BEBT activity, whereas the most similar protein to BEBT in the Arabidopsis proteome does not use benzoyl CoA as a substrate and instead can use acetyl CoA to catalyze the formation of cis-3-hexen-1-yl acetate, a green-leaf volatile.  (+info)

Interaction between visiting bees (Hymenoptera, Apoidea) and flowers of Ludwigia elegans (Camb.) hara (Onagraceae) during the year in two different areas in Sao Paulo, Brazil. (2/13)

This study was designed to characterize the interactions between Ludwigia elegans flowers and visiting bees during two years in two areas 200 km apart, at the same latitude (approximately 22 masculine 48'S) but at different altitudes (Aluminio, 600 m, and Campos do Jordao, 1500 m), in the State of Sao Paulo, Brazil. As these flowers open simultaneously in the morning and lose their petals by sunset, interaction with bees occurs only during the photophase. Flowers of L. elegans were mainly visited by bees, the most frequent species being: Tetraglossula anthracina (Michener, 1989) (Colletidae), Rhophitulus sp. (Andrenidae), and Pseudagapostemon spp. (Halictidae), all considered specialized bees for collecting pollen and nectar from these flowers, as well as the generalist bee Apis mellifera Linnaeus, 1758 (Apidae). The specialist bees were temporally adjusted to the opening schedule of the flower, which occurs primarily in the morning, but shows a circannual variation. T. anthracina appears in both study areas, but only between December and April. The annual activity patterns of these specialist bees are synchronized to the phenology of L. elegans. Photoperiod and temperature cycles are suggested as the main synchronizers of both bees and plants.  (+info)

Sulfate-reducing bacteria in floating macrophyte rhizospheres from an Amazonian floodplain lake in Bolivia and their association with Hg methylation. (3/13)

Five subgroups of sulfate-reducing bacteria (SRB) were detected by PCR in three macrophyte rhizospheres (Polygonum densiflorum, Hymenachne donacifolia, and Ludwigia helminthorriza) and three subgroups in Eichhornia crassipes from La Granja, a floodplain lake from the upper Madeira basin. The SRB community varied according to the macrophyte species but with different degrees of association with their roots. The rhizosphere of the C4 plant Polygonum densiflorum had higher frequencies of SRB subgroups as well as higher mercury methylation potentials (27.5 to 36.1%) and carbon (16.06 +/- 5.40%), nitrogen (2.03 +/- 0.64%), Hg (94.50 +/- 6.86 ng Hg g(-1)), and methylmercury (8.25 +/- 1.45 ng Hg g(-1)) contents than the rhizosphere of the C3 plant Eichhornia crassipes. Mercury methylation in Polygonum densiflorum and Eichhornia crassipes was reduced when SRB metabolism was inhibited by sodium molybdate.  (+info)

Curtobacterium ammoniigenes sp. nov., an ammonia-producing bacterium isolated from plants inhabiting acidic swamps in actual acid sulfate soil areas of Vietnam. (4/13)

The ammonia-producing bacteria B55(T), CA73, SA69 and SA72 were isolated from the waterweeds Ludwigia adscendens (B55(T)) and Eleocharis dulcis (CA73, SA69 and SA72) grown in highly acidic swamps (pH 2-4) in actual acid sulfate soil areas of Vietnam. The isolates were Gram-positive, irregular rod-shaped, non-spore-forming bacteria. On the basis of 16S rRNA gene sequence similarity, strain B55(T) was shown to belong to the genus Curtobacterium of the class Actinobacteria. Chemotaxonomic data (MK-9 as major isoprenoid quinone, d-ornithine as cell-wall diamino acid, acetyl as the acyl type of peptidoglycan) supported the affiliation of all four strains to this genus. Although their 16S rRNA gene sequence similarity was 99 % to species with validly published names within the genus, they formed a group that was distinct in the phylogenetic tree, and DNA-DNA relatedness values to these established species were less than 10 %. The results of physiological and biochemical tests and major fatty acids (cyclohexyl-C(17 : 0), anteiso-C(17 : 0) and cyclohexyl-C(19 : 0)) allowed phenotypic differentiation of these strains from the species of Curtobacterium with validly published names. Therefore, strains B55(T), CA73, SA69 and SA72 represent a novel species, for which the name Curtobacterium ammoniigenes sp. nov. is proposed.
