A new coumestan, tephcalostan (1) has been isolated from the whole plant of Tephrosia calophylla BEDD. together with two known flavonoids, 7-O-methylglabranin (2) and kaempferol 3-O-beta-D-glucopyranoside (3). The structure of tephcalostan was elucidated as 5'-(R)-8, 9-methylenedioxy-5'-isopropenyl-4', 5'-dihydrofurano[2', 3':2, 3]coumestan by extensive one-and two-dimensional (1D- and 2D-)-NMR techniques including (1)H-(1)H correlation spectroscopy (COSY), heteronuclear single quantum coherence (HSQC), heteronuclear multiple bond connectivity (HMBC) and nuclear Overhauser enhancement spectroscopy (NOESY) experiments.  (+info)

Flavonoids, NodD1, NodD2, and nod-box NB15 modulate expression of the y4wEFG locus that is required for indole-3-acetic acid synthesis in Rhizobium sp. strain NGR234. (2/7)

Flavonoids secreted by host plants activate, in conjunction with the transcriptional activator NodD, nod gene expression of rhizobia resulting in the synthesis of Nod factors, which trigger nodule organogenesis. Interestingly, addition of inducing flavonoids also stimulates the production of the phytohormone indole-3-acetic acid (IAA) in several rhizobia. Here, the molecular basis of IAA synthesis in Rhizobium sp. NGR234 was investigated. Mass spectrometric analysis of culture supernatants indicated that NGR234 is capable of synthesizing IAA via three different pathways. The production of IAA is increased strongly by exposure of NGR234 to daidzein in a NodD1-, NodD2-, and SyrM2-dependent manner. This suggests that the y4wEFG locus that is downstream of nod-box NB15 encodes proteins involved in IAA synthesis. Knockout mutations in y4wE and y4wF abolished flavonoid-inducible IAA synthesis and a functional y4wF was required for constitutive IAA production. The promoter activity of NB15 and IAA production both were enhanced by introduction of a multicopy plasmid carrying nodD2 into NGR234. Surprisingly, the y4wE mutant still nodulated Vigna unguiculata and Tephrosia vogelii, although the nodules contained less IAA and IAA conjugates than those formed by the wild-type bacterium.  (+info)

Toxic hepatopathy in sheep associated with the ingestion of the legume Tephrosia cinerea. (3/7)

A disease known as water belly (barriga d'agua), characterized by chronic progressive ascites, affects sheep in the semiarid region of northeastern Brazil. The objectives of this investigation were to study the disease and to determine its cause. Only sheep grazing for long periods in pastures where Tephrosia cinerea represents 80% to 100% of the available forage are affected. Most animals die after a clinical manifestation period of some weeks or months, but others recover when they are withdrawn from the pastures or after the first rains. At necropsy, large amounts of liquid were found in the abdominal cavity, and the liver was hard, with an irregular surface. On histology examination, the main liver lesion was chronic periportal and subcapsular fibrosis with bridging. The disease was produced experimentally in 1 sheep by the administration of large amounts of T. cinerea for 232 days. Another sheep, recovered from the spontaneous disease, had clinical signs after the ingestion of large amounts of the plant for 40 days. Seeds and leaves of the plant were examined by gas chromatography-mass spectrometry for the presence of pyrrolizidine alkaloids with negative results. It is concluded that the disease is caused by the ingestion of T. cinerea.  (+info)

The methanolic extract of the whole plant of Tephrosia pupurea, Linn. was subjected to find out its possible therapeutic utility to validate its folkloric use in native systems of medicine. The extract on application to spontaneous contractions in isolated rabbit jejunum preparations exerted a concentration dependent (0.003-3.0 mg/mL) relaxant effect. The extract also caused concentration dependent relaxation of K+(80 mM)-induced spastic contractions. These findings were further supported by the observations that the extract caused a concentration dependent right ward shift of the Ca2+ response curves in manner similar to that of verapamil. The extract exhibited a relaxant effect on carbachol and high K+ (80 mM)-induced contractions of isolated rabbit tracheal preparations in a manner similar to verapamil. The observed non-specific bronchodilator response is possibly mediated through Ca2+ channel blockade. Moreover, the extract also exhibited a dose dependent relaxant effect on phenylephrine (1 microM) and K+(80 mM)-induced contractions in a manner similar to verapamil. On the basis of the above-mentioned findings, it can be concluded that the use of Tephrosia purpurea, in gastrointestinal spasm, asthma and hypertension is likely to be mediated through calcium channel blockage.  (+info)
