Dietary manipulation to reduce aerial ammonia concentrations in nursery pig facilities. (1/15)
Two 4-wk trials (preliminary study) and three 5-wk trials (major study) were conducted to determine the effects of adding Yucca schidigera extract or anhydrous calcium chloride to nursery diets on the growth performance of nursery pigs and aerial ammonia concentration. The pigs were weaned between 13 and 15 d of age and had an initial BW of 3 to 6 kg. In each trial, pigs were allotted to three identical pig nursery rooms that were environmentally regulated. There were three diets (one diet per room): 1) control, containing 23% CP; 2) control plus 125 ppm of Yucca schidigera extract; and 3) control plus 1.95% anhydrous calcium chloride. Growth performance was recorded weekly. Aerial ammonia concentration was measured daily using aspiration detector tubes and during the last week of each trial using diffusion tubes. Manure samples were collected twice a week during the experimental period to determine ammonia and N concentrations and pH. Plasma urea concentration was determined in blood samples collected from the pigs at the end of each trial. Data were analyzed using split-plot and Latin square designs for the preliminary and major studies, respectively. Feed intake was similar among pigs fed all three diets. There were no differences in ADG and ADG/ADFI (G/F) between pigs fed the control diet and pigs fed the yucca extract diet (P > or = 0.41). In all trials, pigs fed the calcium chloride diet had lower ADG and G/F than pigs fed the other two diets (P < 0.05). In the preliminary study, aerial ammonia tended to be greater in the rooms in which pigs were fed the control diet than in the rooms with pigs fed the yucca extract diet (P = 0.08) and the calcium chloride diet (P = 0.11). In the major study, aerial ammonia increased weekly (diet x week; P < 0.001) in all rooms. In the 4th wk, ammonia concentrations were greater (P < 0.001) in the rooms in which pigs were fed the control diet than in the rooms in which the other two diets were fed. Dietary treatment had no effect on plasma urea concentration (P > or = 0.10), manure ammonia and N concentrations (P > or = 0.50), and manure pH (P > or = 0.78). Although aerial ammonia concentrations were relatively low, the addition of Yucca schidigera extract or calcium chloride to the diet of nursery pigs reduced ammonia concentrations in the nursery rooms. Evolutionary key innovations give organisms access to new ecological resources and cause rapid, sometimes spectacular adaptive radiation. The well known obligate pollination mutualism between yuccas and yucca moths is a major model system for studies of coevolution, and it relies on the key innovation in the moths of complex tentacles used for pollen collecting and active pollination. These structures lack apparent homology in other insects, making them a rare example of a novel limb. We performed anatomical and behavioral studies to determine their origin and found evidence of a remarkably simple mechanism. Morphological analyses of the tentacles and adjacent mouthparts in pollinators and closely related taxa showed that the tentacle appears abruptly in female pollinating yucca moths. Several morphological synapomorphies between the galeae, which constitute the characteristic lepidopteran proboscis, and the tentacle suggest that the tentacle evolved quickly through expression of the genetic template for the galea at an apical growth bud on the first segment of the maxillary palp. Behavioral data indicate that tentacle and proboscis movements are controlled by a shared hydraulic extension mechanism, thus no new mechanism was needed for tentacle function. Known developmental paths from other insects can explain the origin of this sex-specific key innovation in a few steps. The mutagenic activity of yuccaols A, B, and C, trans-resveratrol and trans - 3.3',5.5'-tetrahydroxy -4'-methoxystilbene was tested by the Ames method with Salmonella typhimurium strains TA97, TA98, TA100, TA102 in the absence and presence of metabolic activation (S9 fraction). These phenolic compounds have been isolated and identified from the hark of Yucca schidigera. Plant-insect associations have served as models for investigations of coevolution and the influence of biotic interactions on diversification. The pollination association between yuccas and yucca moths is a classic example of an obligate mutualism often suggested to have been affected by coevolution. Recent work has shown high host specificity in pollinating yucca moths, and here we use Tegeticula yuccasella, the species with the widest diet breadth, to ask how host specificity and isolation by distance contribute to specialization. Isolation by distance at a regional scale was observed in nucleotide variation within the mitochondrial gene cytochrome oxidase I (COI) (r =.294; P =.003). Host-related genetic structure (F(ct) = 0.08) was found to be slightly lower than the level of structure observed between eastern and western moth populations (F(ct) = 0.096). However, 56% of the COI haplotypes sampled from moths on Yucca filamentosa mapped to a host-specific clade in the haplotype network. Taken together, these results suggest that differentiation among T. yuccasella populations on alternative hosts is slight, but gene flow is influenced by both host association and geographic distance. Spectrophotometrically determined substances in Yucca extract, listed as "Yucca foam extract" in the "List of Existing Food Additives in Japan", were investigated by GC/MS, TLC and GC. A TLC method using an anisaldehyde color developing reagent similar to that employed in spectrophotometry was established for selective detection of sapogenins in Yucca extract. Several steroidal sapogenins were found by GC/MS in the fractions corresponding to spots on the TLC plate, and these were assumed to have contributed to the color development in spectrophotometry. Sarsasapogenin and smilagenin were the dominant sapogenins. An on-column injection GC method to determine these sapogenins in Yucca extract was also developed. The sum of these two sapogenins in Yucca extract was 0.9%. The total amount of sapogenin estimated by GC was approximately 2%, which was similar to that measured by spectrophotometry. Mutualisms are balanced antagonistic interactions where both species gain a net benefit. Because mutualisms generate resources, they can be exploited by individuals that reap the benefits of the interaction without paying any cost. The presence of such 'cheaters' may have important consequences, yet we are only beginning to understand how cheaters evolve from mutualists and how their evolution may be curtailed within mutualistic lineages. The yucca-yucca moth pollination mutualism is an excellent model in this context as there have been two origins of cheating from within the yucca moth lineage. We used nuclear and mitochondrial DNA markers to examine genetic structure in a moth population where a cheater species is parapatric with a resident pollinator. The results revealed extensive hybridization between pollinators and cheaters. Hybrids were genetically intermediate to parental populations, even though all individuals in this population had a pollinator phenotype. The results suggest that mutualisms can be stable in the face of introgression of cheater genes and that the ability of cheaters to invade a given mutualism may be more limited than previously appreciated. The yucca-yucca moth interaction is one of the most well-known and remarkable obligate pollination mutualisms, and is an important study system for understanding coevolution. Previous research suggests that specialist pollinators can promote rapid diversification in plants, and theoretical work has predicted that obligate pollination mutualism promotes cospeciation between plants and their pollinators, resulting in contemporaneous, parallel diversification. However, a lack of information about the age of Yucca has impeded efforts to test these hypotheses. We used analyses of 4322 AFLP markers and cpDNA sequence data representing six non-protein-coding regions (trnT-trnL, trnL, trnL intron, trnL-trnF, rps16 and clpP intron 2) from all 34 species to recover a consensus organismal phylogeny, and used penalized likelihood to estimate divergence times and speciation rates in Yucca. The results indicate that the pollination mutualism did not accelerate diversification, as Yucca diversity (34 species) is not significantly greater than that of its non-moth-pollinated sister group, Agave sensu latissimus (240 species). The new phylogenetic estimates also corroborate the suggestion that the plant-moth pollination mutualism has at least two origins within the Agavaceae. Finally, age estimates show significant discord between the age of Yucca (ca 6-10Myr) and the current best estimates for the age of their pollinators (32-40Myr). This study explored the occurrence and distribution of myxomycete species on the aerial reproductive structures of vascular plants. Eight species of vascular plants representing five families were sampled. The doubled rope climbing method was used to collect bark and cones from the canopy of Pinus echinata. Bark and aerial seed pods were gathered from Cercis canadensis, follicles and stems from Asclepias syriaca, dried composite inflorescences and stems from Echinacea angustifolia, E. pallida, and E. paradoxa var. paradoxa, and capsules and stems from Yucca glauca and Y. smalliana. Reproductive structures and bark/stems for 202 host plants were separated and cultured in 541 moist chambers, resulting in 118 collections yielding 32 myxomycete species representing 11 genera, seven families and five orders. There was no significant difference in pH values of the reproductive structures and bark/stems of the host plants, however legume pods of C. canadensis (6.9 +/- 1.3) had higher pH than the bark (6.0 +/- 1.1) and had a different composition of myxomycete species. Myxomycete orders have optimal pH ranges. Nonmetric multidimensional scaling, multiresponse permutation procedure and indicator species analysis showed a significant difference in species richness of reproductive structures and bark/stems. The bark of trees had greater mean species richness of myxomycetes than the reproductive structures, but the reproductive structures of herbaceous plants had greater mean species richness of myxomycetes than the stems. A new term, herbicolous myxomycetes, is proposed for a group of myxomycetes frequently associated with herbaceous, perennial, grassland plants.
