The chloroplast genome of Nymphaea alba: whole-genome analyses and the problem of identifying the most basal angiosperm. (1/14)
Angiosperms (flowering plants) dominate contemporary terrestrial flora with roughly 250,000 species, but their origin and early evolution are still poorly understood. In recent years, molecular evidence has accumulated suggesting a dicotyledonous origin of monocots. Phylogenetic reconstructions have suggested that several dicotyledonous groups that include taxa such as Amborella, Austrobaileya, and Nymphaea branch off as the most basal among angiosperms. This has led to the concept of monocots, "eudicots," "basal dicots," and "ANITA" groupings. Here, we present the sequence and phylogenetic analyses of the chloroplast DNA of Nymphaea alba. Phylogenetic analyses of our 14-species data set, consisting of 29,991 aligned nucleotide positions per chloroplast genome, revealed consistent support for Nymphaea being a divergent member of a monophyletic dicot assemblage. Three distinct angiosperm lineages were supported in the majority of our phylogenetic analyses-eudicots, Magnoliopsida, and monocots. However, the monocot lineage leading to the grasses was the deepest branching. Although analyses of only one individual gene alignment (out of 61) is consistent with some recently proposed hypotheses for the paraphyly of dicots, we also report observations that nine genes do not support paraphyly of dicots. Instead, they support the basal monocot-dicot split. Consistent with this finding, we also report observations suggesting that the monocot lineage leading to the grasses has the strongest phylogenetic affinity to gymnosperms. Our findings have general implications for studies of substitution model specification and analyses of concatenated genome data. BACKGROUND AND AIMS: Several families of tropical plants have thermogenic flowers that show a 2-d protogynous sequence. Most are pollinated by large beetles that remain for the entire period in the flowers, where they compete for mates and feed. Active beetles require high body temperatures that they can achieve endogenously at great energy expense or attain passively and cheaply in a warm environment. Floral heating is therefore hypothesized to be a direct energy reward to endothermic beetles, in addition to its accepted role in enhancing scent production. METHODS: This study measures the pattern of floral heat production (as temperature in 20 flowers and respiration rates in five flowers) in Victoria amazonica at field sites in Guyana and correlates floral temperatures with body temperatures necessary for activity in visiting Cyclocephala hardyi beetles. KEY RESULTS: Thermogenesis occurred in a bimodal pattern, with peaks associated with the arrival and departure of beetles near sunset. Peak CO(2) production rates averaged 2.9 micromol s(-1), equivalent to a heat production of 1.4 W. Heat was generated mainly in the floral chamber on the first evening and by the stamen complex on the second. Mean chamber temperature remained between 29.3 and 34.7 degrees C during the first night, when ambient temperature was 23.5-25.2 degrees C. Beetles actively competed for mates and consumed stylar processes in the floral chamber, where their mean thoracic temperature was 33.2 degrees C. At the lower ambient temperatures outside of the flower, beetles capable of sustained flight had a similar mean temperature of 32.0 degrees C. CONCLUSIONS: Floral heating is not only associated with attraction, but continues throughout the night when beetles are active inside the flower and increases again when they leave. Floral chamber temperatures similar to activity temperatures of actively endothermic beetles imply that thermogenesis is an energy reward. INTRODUCTION: This study aims to investigate Nymphaea stellata (N. stellata) flower extract for antihyperglycaemic and antihyperlipidaemic effects in diabetic rats induced by alloxan. Its effect was compared with that of glibenclamide, a reference antidiabetic drug. METHODS: Diabetic animals were randomly divided into five groups and treated orally with different doses (200, 300 and 400 mg/kg body weight) of flower extract once a day for 30 days. The body weight of each animal was determined, to assess any possible weight gain or loss in experimental animals compared with control groups. On the 31st day, those administered 300 mg/kg of N. stellata flower showed more promising results with regard to fasting blood glucose (FBG), plasma insulin levels, haemoglobin counts, urine sugar levels, food intake, water intake, urea and protein when compared to those treated with other doses. Therefore, 300 mg/kg dose was used for further biochemical studies. Total lipids (TL), total cholesterol (TC), triglycerides (TG), phospholipids, free fatty acids (FFA), low density lipoproteins (LDL), very low density lipoproteins (VLDL), atherogenic index (AI) and high density lipoproteins (HDL) levels, on normal and diabetic rats treated with the dose of 300 mg/kg, were evaluated. RESULTS: The flower extract shows a significant (p-value is less than 0.001) reduction in levels of FBG, water intake, food intake, urine sugar, blood urea, TL, TC, TG, FFA, phospholipids, LDL, VLDL and AI. It also shows a significant increase in body weight, plasma insulin, protein, haemoglobin and HDL levels. CONCLUSION: Our results suggest that N. stellata flower extract exhibit antihyperglycaemic as well as antihyperlipidaemic effects on alloxan-induced diabetic rats. Alascospora evergladensis, a freshwater ascomycete collected from submerged dead petioles of Nymphaea odorata during a survey of aquatic fungi along a phosphorus gradient in the Florida Everglades, is described and illustrated as a new genus and species in the Pleosporales (Pleosporomycetidae, Dothideomycetes). The new fungus is unique among genera in the Pleosporales based on a combination of morphological characters that include light brown, translucent, membranous, ostiolate ascomata with dark, amorphous material irregularly deposited on the peridium, especially around the ostiole; globose, fissitunicate, thick-walled asci; septate pseudoparaphyses; and 1-septate ascospores that are hyaline when young, and surrounded by a hyaline gelatinous sheath that is wing-shaped in outline on each side of the ascospore.
