In particular, different views on the centrality of population growth rates to the study of population dynamics and the role of experiments and theory are explored. Major themes emerging include the role of modern statistical techniques in bringing together experimental and theoretical studies, the importance of long-term experimentation and the need for ecology to have model systems, and the value of population growth rate as a means of understanding and predicting population change. Genetic diversity in Primula obconica (Primulaceae) from central and south-west China as revealed by ISSR markers. (2/24)

Genetic diversity was investigated in 60 individuals of Primula obconica from four natural populations (from Hubei, central China, and Sichuan, south-west China) and from cultivated material. Inter-simple sequence repeat (ISSR) techniques produced 249 polymorphic bands and identified 60 ISSR genotypes. Based on Shannon's index and Nei's genetic diversity, the genetic diversity detected in all natural populations of P. obconica was much higher than that in the cultivated plants, and that in the three Dalaoling (Hubei) populations was higher than that in the Wawushan (Sichuan) population. UPGMA cluster analysis showed that there was no distinct genetic differentiation between populations from the Mt Dalaoling area and the Mt Wawushan area. This study provides a population-level genetic profile of P. obconica, which was previously poorly known but which is important for Primula breeding and cultivation.  (+info)

Identification of primula fatty acid delta 6-desaturases with n-3 substrate preferences. (3/24)

Fatty acid Delta(6)-desaturation, the first committed step in C(20) polyunsaturated fatty acid biosynthesis, is generally considered not to discriminate between n-3 and n-6 substrates. We previously identified higher plant species that showed preferential Delta(6)-desaturation of n-3 C(18) fatty acid substrates. A polymerase chain reaction-based approach was used to isolate 'front-end' cytochrome b(5) fusion desaturases from Primula vialii Franchet and Primula farinosa L. Functional analysis in Saccharomyces cerevisiae identified fatty acid Delta(6)-desaturases with a strong specificity for the n-3 substrate alpha-linolenic acid (18:3 Delta(9,12,15)). These results indicate that the accumulation of octadecatetraenoic acid (18:4 Delta(6,9,12,15)) in planta is due to the activity of a novel n-3-specific fatty acid Delta(6)-desaturase.  (+info)

Analysis of petal shape variation of Primula sieboldii by elliptic fourier descriptors and principal component analysis. (4/24)

BACKGROUND AND AIMS: Petals are important for Primula sieboldii because of the commercial value of its flowers, and their form is a target characteristic for breeding. An appropriate understanding of petal form in terms of genetic mechanisms and environmental effects is necessary for improvement of this species. The aim of this study was to establish a quantitative evaluation method of petal shape by elliptic Fourier descriptors and principal component analysis (EF-PCA), and thus to investigate genotypic and environmental effects on petal morphology. METHODS: EF-PCA describes an overall shape mathematically by transforming coordinate information concerning its contours into elliptic Fourier descriptors (EFDs) and summarizing the EFDs by principal component analysis. To examine varietal effects on principal component (PC) scores and petal area among commercial varieties, nested ANOVAs were performed (since the samples had a hierarchical structure with four sources, i.e. variety, plant, flower and petal). KEY RESULTS: Petal shape variation could be evaluated successfully and the symmetrical and asymmetrical elements of the overall shape variation could be detected. The proportions of the variance component due to varietal differences were more than 70 % in the first five PCs of the symmetrical elements and petal area. By contrast, the proportions due to varietal effects of all PCs of the asymmetrical elements were less than 20 %, and the proportions of the variation within a flower were more than 75 %. It was also demonstrated that the yearly variance of petal shape was small, and that of petal area was large. CONCLUSIONS: Within a flower the major source of the symmetrical elements is genotypic and the asymmetrical elements are strongly affected by the environment. With respect to petal area, the contribution of genotypes is also large; it is, however, affected by the macro-environment more notably than is petal shape.  (+info)

Molecular characterization of DNA sequences from the Primula vulgaris S-locus. (5/24)

Primula species provide possibly the best known examples of heteromorphic flower development and this breeding system has attracted considerable attention, including that of Charles Darwin. However, despite considerable recent advances in molecular genetics, nothing is known about the molecular basis of floral heteromorphy. The first molecular marker for the Primula S-locus is reported here. This DNA sequence was identified by random amplification of polymorphic DNA (RAPD)-PCR, further defined as a sequence characterized amplified region (SCAR) marker, and subsequently shown to correspond to a restriction fragment length polymorphism (RFLP) that is linked to the thrum allele of the Primula S-locus. The sequence of 8.8 kb of genomic DNA encompassing this thrum-specific RFLP is presented. Analysis of this DNA reveals a highly repetitive sequence structure similar to that found at the S-locus in other species; it also contains sequences similar to elements of a Gypsy-like retrotransposon. The identification of a specific DNA sequence associated with the thrum allele of the Primula S-locus provides the first molecular probe with which to investigate the molecular basis of heteromorphic flower development in Primula.  (+info)

Quantitative evaluation of the petal shape variation in Primula sieboldii caused by breeding process in the last 300 years. (6/24)

Primula sieboldii: (E. Morren) has been a popular garden plant at least since the Edo period, about 300 years ago. We compared petal form between cultivars and wild populations in order to characterise the changes that have occurred during domestication. The comparison was made using EF-PCA analysis, which describes overall petal shape mathematically by transforming petal contour coordinates into elliptic Fourier descriptors; it subsequently summarises these descriptors by principal component analysis (PCA). Rearing cultivars in a common-garden experiment identified the PCs with a substantial genetic element. A clear heritable component was detected for the PCs characterising symmetrical variation in flower shape, but not the asymmetrical variation. Wild populations of this species have become endangered owing to habitat destruction by human activity, and many lowland floodplain habitats have been lost. Variation within the remaining wild populations was significantly lower than in the cultivars for PC1 (aspect ratio), PC3 (curvature of proximal and distal parts) and petal area; but not for PC2 (depth of head notch) and PC4 (position of the centre of gravity). The shifts in petal form from the wild populations to the cultivars parallel those seen in other crop-types following domestication, including an increase in size and diversity of forms: cultivars have shallower head notches, more fan-shaped petals and larger petals than do wild P. sieboldii.  (+info)

Ability of bumblebees to discriminate differences in the shape of artificial flowers of Primula sieboldii (Primulaceae). (7/24)

BACKGROUND AND AIMS: Flower shapes are important visual cues for pollinators. However, the ability of pollinators to discriminate between flower shapes under natural conditions is poorly understood. This study focused on the diversity of flower shape in Primula sieboldii and investigated the ability of bumblebees to discriminate between flowers by combining computer graphics with a traditional behavioural experiment. METHODS: Elliptic Fourier descriptors described shapes by transforming coordinate information for the contours into coefficients, and principal components analysis summarized these coefficients. Using these methods, artificial flowers were created based on the natural diversity of petal shape in P. sieboldii. Dual-choice tests were then performed to investigate the ability of the bumblebees to detect differences in the aspect ratio of petals and the depth of their head notch. KEY RESULTS: The insects showed no significant ability to detect differences in the aspect ratio of the petals under natural conditions unless the morphological distance increased to an unrealistic level. These results suggest the existence of a perception threshold for distances in this parameter. The bumblebees showed a significant preference for narrow petals even after training using flowers with wide petals. The bumblebees showed a significant ability to discriminate based on the depth of the petal head notch after training using artificial flowers with a deep head notch. However, they showed no discrimination in tests with training using extreme distances between flowers in this parameter. CONCLUSIONS: A new type of behavioural experiment was demonstrated using real variation in flower corolla shape in P. sieboldii. If the range in aspect ratios of petals expands much further, bumblebees may learn to exhibit selective behaviour. However, because discrimination by bumblebees under natural conditions was low, there may be no strong selective behaviour based on innate or learned preferences under natural conditions.  (+info)
