Ultracytochemical study of medullary bone calcification in estrogen injected male Japanese quail. (1/9)

Fine structural and cytochemical studies were performed to clarify the pattern of medullary bone calcification, specifically in relation to sulfated glycosaminoglycans, by using estrogen-induced medullary bone of male Japanese quails. Tibiae were collected at 4 and 7 days after estrogen treatment. Medullary bone had developed inward toward the marrow cavity, and calcification had begun near the cortical bone and deeper parts of the trabeculae, accompanied by wide osteoid at extending tips and surface areas of the trabeculae. Sulfated glycosaminoglycans, detected by high iron diamine (HID), were distributed in the matrix in a pattern similar to that of calcified matrix of the trabeculae. Cortical bone was negatively stained by HID. In undecalcified specimens, calcified nodules were seen in areas undergoing calcification. Globular structures composed of fine filamentous materials, a marginal dense layer, and central core, were also observed in the matrix of decalcified specimens. Both the calcified nodules and globular structures showed the same distribution pattern, i.e., they were dispersed at surface areas and coalesced in the deeper areas of the matrix. The globular structures were exclusively positive for HID-thiocarbohydrazide-silver protein (HID-TCH-SP) stain, indicating the localization of sulfated glycosaminoglycans. These results strongly suggest that medullary bone calcification progresses by the coalescence of calcified nodules and that sulfated glycosaminoglycans play an important role for the regulation of globular calcification.  (+info)

Seromucous cells in human sublingual glands: examination by immunocytochemistry of lysozyme. (2/9)

This study examined the occurrence and morphological features of serous-type cells in human sublingual gland, using immunocytochemistry for lysozyme. Lysozyme-positive cells usually formed demilunes and occasionally their own acini. They were also found among cells of an intercalated duct and in its immature acinus consisting of a small number of secretory cells. All these serous cells could be classified as seromucous cells because they simultaneously revealed reactivity for mucus, i.e., a periodic acid-Schiff (PAS) and a periodic acid-thiocarbohydrazide-silver proteinate (PA-TCHSP) reaction under the light- and electron-microscope, respectively. Immunogold labeling of lysozyme in the seromucous cells was distributed on variously sized secretory granules. These usually possessed a single electron-dense spherule in an electron-lucent matrix, while granules of a homogenous structure were also present. Lysozyme-positive cells filled with large, lucent secretory granules could hardly be morphologically distinguished from the lysozyme-negative mucous cells; they corresponded to "intermediate" cells designated under the light microscope. All "immature" secretory cells with only a few secretory granules were also lysozyme-positive seromucous cells. The present study demonstrated that the seromucous cells in the human sublingual glands conform closely with those in the human labial glands (MIYAZAKI et. al., 1998).  (+info)

Detection of silver protein complex injections in the bovine udder using X-ray fluorescence: a preliminary investigation. (3/9)

To determine the feasibility of using x-ray fluorescence (XRF) to detect the presence of silver in the mammary gland of dairy cows injected with mild silver protein suspension. The XRF spectroscopy was conducted on cadaver udders with and without mild silver protein injected. Spectral analysis was performed in order to determine the amplitude of the silver K-alpha peak that was detected. By comparing the amplitude of the K-alpha peak to the background, a minimum time of collection was determined, as a measure of the time required to observe a silver signal that is significantly non-zero. The minimum detection time required for evidence of injected silver suspension was calculated to be 2.8 +/- 0.2 s. Even with an additional requirement that the net signal exceed 50 counts, the clear indication of the presence of silver will be observed within 4 min of interrogation. X-ray fluorescence spectroscopy was shown to be a viable method for the detection of injected silver protein in cadaver mammary glands of dairy cows. While these findings are promising, further studies must be conducted to investigate the time dependence of the silver signal when diffusion, absorption, and redistribution are involved, under conditions that better mimic those encountered at an exhibition. This technique, used in conjunction with screening ultrasound examinations, has the potential to confirm sites of injection of silver compounds.  (+info)

Immunological evidence for a relationship between the dendrotoxin-binding protein and the mammalian homologue of the Drosophila Shaker K+ channel. (5/9)

Polyclonal antibodies were raised against two synthetic peptides from different parts of the predicted amino acid sequence of the mouse homologue (MBK1) of the Drosophila Shaker K+ channel. The antibodies recognized the toxin-binding subunit of the dendrotoxin-binding proteins from rat and bovine brain. The results suggest that the dendrotoxin-binding protein is related to the expression products of the mammalian homologue of the Shaker gene.  (+info)

Ins(1,4,5)P3 5-phosphatase catalyzes the dephosphorylation of Ins(1,4,5)P3 in the 5-position. In a high speed soluble fraction of bovine brain, there are two soluble 5-phosphatases: type I and type II. The purified Ins(1,4,5)P3 5-phosphatase type I exhibits a major silver-stained band of 43 kDa on denaturing (SDS) gels. It is possible to extract the 5-phosphatase activity form a duplicate lane after gel electrophoresis. The 43 kDa region contains the extractable Ins(1,4,5)P3 5-phosphatase activity.  (+info)

Characterization of the phosphorylation sites in the chicken and bovine myristoylated alanine-rich C kinase substrate protein, a prominent cellular substrate for protein kinase C. (7/9)

Little is known about the important cellular substrates for protein kinase C and their potential roles in mediating protein kinase C-dependent processes. We evaluated the protein kinase C phosphorylation sites in a major cellular substrate for the kinase, a protein of apparent Mr 80,000 in bovine and 60,000 in chicken tissues; we have recently determined the primary sequences of these proteins and tentatively named them the myristoylated alanine-rich C kinase substrates. The proteins were purified to apparent homogeneity from bovine and chicken brains, phosphorylated with protein kinase C, digested with trypsin, and the phosphopeptides purified and sequenced. Four distinct phosphopeptides were identified from both the bovine and chicken proteins. Two of the phosphorylated serines were contained in the repeated motif FSFKK, one in the sequence LSGF, and one in the sequence SFK. All four sites were contained within a basic domain of 25 amino acids which was identical in the chicken and bovine proteins. All of the sites phosphorylated in the cell-free system appeared to be phosphorylated in intact cells; an additional site may have been present in the proteins from intact cells. The identity of the phosphorylation site domains from two proteins of overall 65% amino acid sequence identity suggests a potential role for this domain in the physiological function of the myristoylated alanine-rich C kinase substrate proteins.  (+info)

Cytochemical localization of glycogen in Chlamydia trachomatis inclusions. (8/9)

The origin and distribution of glycogen in inclusions of Chlamydia trachomatis were demonstrated with silver proteinate stain for electron microscopy. Glycogen particles were detected in all developmental stages of C. trachomatis, as well as free in the inclusions. Intrachlamydial glycogen was most common in elementary bodies but was also detected in intermediate forms and reticulate bodies (RBs). Abnormal divisions and breakdown of cytoplasmic membranes were common in RBs. Cytoplasmic contents, including glycogen particles, were released into the inclusions after rupture of the outer membranes of abnormal RBs and intermediate forms. From these observations, we conclude that glycogen in inclusions of C. trachomatis originates in the organisms themselves.  (+info)
